One Health-oriented research progress on biological recognition of emerging organic contaminants

摘要
     Guided by the national Soil Pollution Prevention and Control Action Plan, this study focuses on the pollution caused by ubiquitous emerging contaminants in soil and systematically investigates their environmental behaviors and risk identification. Rapid and accurate detection of emerging contaminants in soil constitutes a critical link in preventing and controlling their pollution risks. Current detection methods for emerging contaminants suffer from numerous drawbacks, including high consumption of organic reagents, cumbersome extraction procedures and low detection throughput. Furthermore, their test results fail to truly reflect the biological hazards posed by emerging contaminants.
     This research integrates metagenomics and biosensor technologies to mine genetic elements regulated and induced by emerging contaminants from the metagenomes of contaminated samples. Molecular biological techniques are adopted to construct whole-cell biosensors capable of responding to concentration variations of emerging contaminants. Combined analytical chemistry and microbiological approaches are employed to explore extraction protocols and detection workflows for extractable emerging contaminants in soil, as well as their correlations with bioavailability. Portable biological test strips targeting emerging contaminants are fabricated, and a workflow for semi-quantitative detection of emerging contaminants in field contaminated soil is established. The developed method is applied to determine the contamination levels of emerging contaminants in soil samples collected from polluted sites, and the detection performance of the assay toward soil-borne emerging contaminants is analyzed. Ultimately, quantitative and semi-quantitative detection methodologies for emerging contaminants in soil are established.
  The detection methods developed in this project facilitate the elucidation of the distribution and actual hazard profiles of emerging contaminants, and provide a scientific basis for formulating targeted strategies for the precise prevention and control of emerging contaminant pollution.
